Affinity optimizing enhancer variants disrupt development

Talk abstract: An emerging regulatory principle governing enhancers is the use of suboptimal affinity binding sites to encode tissue-specific gene expression during development. Here we discover that affinity optimizing single-nucleotide variants can violate this principle, disrupt tissue-specific gene expression, and alter phenotypes in Ciona, mice, and humans.
